چکیده

N atural ageing of the skin is enhanced by environmental factors. Solar UV exposure is one of the main exogenous factors that is participating in that process and that can lead to photoageing, immunosuppression, and photocarcinogenesis. Using a new RSF method to determine UVinduced free radical injuries in the skin, it is possible to quantify the protective effect of sunscreens and UV-filters. The method is based on Electron Spin Resonance (ESR) spectroscopy measurements. Induction of free radicals in skin depends on the wavelength and dose of UV radiation. UVA penetrates into the deeper layers of the skin and induces more ROS as compared with UVB radiation. Therefore, UV filters that absorb or scatter UVA radiation give a stronger protection against free radical damage. In this study different sunscreen formulations have been tested in order to quantify their protection against free radicals. Abstract Both UVA and UVB irradiation are very damaging to the skin. Depending on the wavelength, UV damage occurs via different mechanisms. UVA mainly produces free radicals (FR)/reactive oxygen species (ROS) though interaction with endogenous photosensitizers. These ROS will cause indirect damage to DNA, proteins and membranes. ROS is believed to be involved in photodamage of dermal connective tissue cells and proteins. On the contrary, DNA with its aromatic, heterocyclic bases is a strongly absorbing chromophore for UVB (absorption maximum at 260-265nm). Direct absorption of the UVB photons leads to disruption of DNA, Introduction

UV-generated free radicals (FR) in skin: their prevention by sunscreens and their induction by self-tanning agents.

In the past few years, the cellular effects of ultraviolet (UV) irradiation induced in skin have become increasingly recognized. Indeed, it is now well known that UV irradiation induces structural and cellular changes in all the compartments of skin tissue. The generation of reactive oxygen species (ROS) is the first and immediate consequence of UV exposure and therefore the quantitative determ...

Measurements of UV-generated free radicals/reactive oxygen species (ROS) in skin.

Free radicals/reactive oxygen species (ROS) generated in skin by UV irradiation were measured by electron spin resonance (ESR). To increase the sensitivity of measurement the short life free radicals/ROS were scavenged and accumulated by using the nitroxyl probe 3-carboxy-2,2,5,5-tetrametylpyrrolidine-1-oxyl (PCA). The spatial distribution of free radicals/ROS measured in pig skin biopsies with...

Skin protection against ultraviolet induced free radicals with ascorbyl palmitate in microemulsions.

UV irradiation induces free radical formation in the skin. UV filters and antioxidants can be used for protection. In the present work, the amphiphilic antioxidant ascorbyl palmitate has been investigated and its effectiveness against free radical formation in porcine skin determined with electron paramagnetic resonance (EPR) spectroscopy with a spin trapping technique. 5-(diethoxyphosphoryl)-5...

Light - Instead of UV Protection: New Requirements for Skin Cancer Prevention.

The requirements on sunscreens have essentially changed, since some years ago it was demonstrated that approximately 50% of free radicals, that are formed in the skin by solar radiation, originate from the visible and infrared regions of the solar spectrum. In addition, a critical radical concentration threshold could be found. If this concentration, the free radical threshold value (FRTV), is ...

Correlation Between Sun Protection Factor and Antioxidant Activity, Phenol and Flavonoid Contents of Some Medicinal Plants

Long exposure of UV radiation increases risk of skin diseases such as cancer and photoallergic reactions. UV-B (280-320 nm) radiation is mainly responsible for inducing skin problems. Skin protection is a suitable method in the fight against ultraviolet radiation induced damage. Various synthetic agents have been used as photo protective but because of their potential toxicity in humans, they h...
